چکیده

1. Photosynthetic electron transport from water to lipophilic Photosystem II acceptors was stimulated 3--5-fold by high concentrations (greater than or equal to 1 M) of salts containing anions such as citrate, succinate and phosphate that are high in the Hofmeister series. 2. In trypsin-treated chloroplasts, K3Fe(CN)6 reduction insensitive to 3-(3,4-dichlorophenyl)-1,1-dimethylurea was strongly stimulated by high concentrations of potassium citrate, but there was much less stimulation of 2,6-dichloroindophenol reduction in Tris-treated chloroplasts supplied with 1,5-diphenylcarbazide as artificial donor. The results suggest that the main site of action of citrate was the O2-evolving complex of Photosystem II. 3. Photosystem I partial reactions were also stimulated by intermediate concentrations of citrate (up to 2-fold stimulation by 0.6--0.8 M-citrate), but were inhibited at the highest concentrations. The observed stimulation may have been caused by stabilizaton of plastocyanin that was complexed with the Photosystem I reaction centre, 4. At 1 M, potassium citrate protected O2 evolution against denaturation by heat or by the chaotropic agent NaNO3. 5. It is suggested that anions high in the Hofmeister series stimulated and stabilized electron transport by enhancing water structure around the protein complexes in the thylakoid membrane.

Biochar impact on physiological and biochemical attributes of Spinach (Spinacia oleracea L.) in nickel contaminated soil

Disastrous effect of nickel on spinach was discussed by number of authors but the effect of amendments like biochar with nickel on Spinacea oleraceaL. is not still discussed by any author of the world because biochar was used as soil amendments which play a vital role in reducing mobilization and uptake of nickel by spinach plants.  As nickel contaminated plants are very harmful for th...

Inhibition of photosynthetic carbon dioxide fixation in isolated spinach chloroplasts exposed to reduced osmotic potentials.

Reduced osmotic potentials inhibited the rate of CO(2) fixation by isolated intact spinach (Spinacia oleracea) chloroplasts. This inhibition was observed immediately after transfer of chloroplasts from a solution containing 0.33 m sorbitol to higher sorbitol concentrations, and the depressed rate remained constant. The inhibited CO(2) fixation could not be attributed to a decreased rate of phot...

Light-Dependent Conversions of Endogenous alpha-Tocopherylquinone and Plastoquinone-D in Spinacia Oleracea Chloroplasts.

Plastoquinone, a substituted benzoquinone occurring in the chloroplasts of plants (5), has been shown to be involved in and required for various photosynthetic electron transport reactions. Bishop (2) has shown that PQ-A4 is required to restore photoreductive activity to petroleum ether extracted chloroplasts. Similarly Krogmann (15) demonstrated the requirement for PQ-A in photophosphorylation...

Carbon dioxide fixation in the light and in the dark by isolated spinach chloroplasts.

Factors affecting CO(2) fixation in the spinach (Spinacia oleracea) chloroplast were investigated. Free magnesium ions are shown to be highly inhibitory for photosynthetic CO(2) fixation in isolated intact spinach chloroplasts. The pH optimum for CO(2) fixation is about 8.5 but is dependent upon the reaction medium. Conditions are defined under which chloroplasts illuminated in the absence of C...

Regulation of Photosynthetic Electron Transport in Intact Spinach Chloroplasts: I. INFLUENCE OF EXOGENOUS SALTS ON OXALOACETATE REDUCTION.

Relatively high concentrations of monovalent salts (150 millimolar) stimulated light-saturated uncoupled rates of O(2) evolution linked to oxaloacetic acid (OAA) reduction by intact chloroplasts 2-to 3-fold. In contrast, monovalent salts partially inhibited light-saturated rates of O(2) evolution coupled to CO(2) fixation and uncoupled rates of nitrite reduction. In the presence of high salt co...
